TS2 and TS3 Thunderbolt docks as well as FASTA-6GU3 Plus card are with "Command Based" Port Multiplier SATA Host controllers and can support up to 8 Hard Disks or SSDs in total from two Port Multiplier SATA devices behind TS2 and TS3.
For example, CalDigit VR is a 2-Drive Port Multiplier box. If you plugged-in 2x CalDigit VRs (in RAID 0 mode) to TS2's eSATA ports, you will see both VRs in Windows or macOS.
If your SATA Port Multiplier box requires an FIS-based controller to maintain multiple disks in RAID or JBOD mode, the TS2 / TS3 / Plus card may not be able to recognize your FIS-based box OR the performance may be significantly slower than plugging-in to the FIS-based host controller.
On Windows 10 platform, the in-box SATA driver is ready to support Plus card's, TS2's and TS3's eSATA Ports, however, if you find any problem when connecting to Port Multiplier boxes, please contact Support@CalDigit.com to obtain the latest SATA driver.
